The "Muppets Tonight Theme" opened every episode of the series. It features Clifford as lead singer, while the camera follows the KMUP studio activity into the control room, dressing area, and elsewhere as the Muppets prepare for the show. Jowls of the Muppets Tonight Band has a whistling solo.
An instrumental version of the song appears over the end credits, initially over a still shot of the KMUP exterior for the first five episodes. For subsequent episodes, the music underscored outtakes.

- The theme is preceded by a spoken The Muppet Show style introduction (complete with its own theme melody as a short musical cue) by Kermit the Frog in front of the studio.
- In the first season, a black and white photo of the Pigs in Space cast could be seen in the background behind Kermit (replaced by a Muppets Tonight cast photo for the second season).
