"Toothy Takes a Partner" refers to a 1978 Sesame Street sketch and the song used in it.
Toothy, a set of teeth (Jerry Nelson), bemoans to his agent Manny (Jim Henson) that he needs a new partner for his act, which has lost its shine. Manny calls in some potential partners - a Washcloth and Hairbrush, which don't fit the bill. Finally, a Toothbrush arrives and Toothy is instantly struck by her. Her friend, Pastey (Frank Oz), applies some toothpaste to her bristles and she cleans up Toothy. The new act performs a musical number in celebration of their new partnership.
